Faktor nekrosis tumor-alfa

Faktor nekrosis tumor-α (bahasa Inggris: Tumor necrosis factor alpha, cachexin, cachectin, DIF; TNFA, TNFSF2, TNF-alpha, TNF-α) adalah sitokina yang banyak disekresi oleh makrofaga dan memiliki banyak peran metabolisme seperti proliferasi sel, differensiasi, apoptosis, metabolisme lipid, dan koagulasi.[1]

TNF-α merupakan anggota dari keluarga faktor nekrosis tumor yang memiliki pencerap seperti TNFRSF1A/TNFR1 dan TNFRSF1B/TNFBR.

Meskipun TNF-α menginduksi migrasi sel punca mesenkimal ke tempat yang diperlukan dengan mekanisme TRAIL,[2] rasio plasma TNF-α yang tinggi menunjukkan disfungsi miokardial pasca-iskemia, sindrom metabolisme, gangguan laju filtrasi glomerular, rasio asam urat yang tinggi, rendahnya kadar HDL,[3] dan bersama IL-6 dapat menjadi faktor pemicu gagal jantung atau Left ventricular diastolic dysfunction.[4] Pada hepatosit, TNF-α akan meningkatkan kadar SCAP di dalam sitoplasma dan menstimulasi sintesis trigliserida, sehingga TNF-α dianggap berperan dalam patogenesis steatosis.[5]

Adenosina,[3] kurkumin,[6] merupakan penghambat TNF-α.
